Most pipettes work on the principle of creating a vacuum by reducing the volume of the pipette or pipetting bulb, thereby expelling the air from the pipette. After the pipette is inserted into a liquid, the internal volume within the pipette is slowly increased back to its original value. This increase in volume creates a suction mechanism. As there is no more air available, a quantity of liquid is sucked in that is approximately equal to the amount of air lost. By then reducing the volume again, the liquid is forced out of the pipette.
